Experimental Lung‑Disease Drug Shows Early Signs of Slowing Biological Aging

Fort Wayne, IN – Researchers at Insilico Medicine reported that rentosertib, an experimental medication being tested for idiopathic pulmonary fibrosis (IPF), may have an unexpected benefit: it appears to make patients biologically younger on several established aging measures.

Study design and key findings

The pilot study enrolled 42 patients with IPF, a serious condition that causes progressive scarring of lung tissue. Participants received rentosertib at a dose of 30 mg twice daily for four weeks, after which blood samples were analyzed for protein patterns linked to biological age.

Six independent biological‑aging clocks showed that patients on rentosertib registered as younger than those receiving a placebo. Researchers estimated the effect corresponded to roughly three to four years of reduced biological age for most participants, with one individual showing a six‑year difference.

Interpretation and cautions

Lead author Alex Zhavoronkov, Ph.D., emphasized that the findings do not prove the drug can reverse aging or extend lifespan. “It basically means that you are predicted to be younger using a validated set of aging clocks. We don’t know what that means yet,” he said.

Because IPF itself can alter many of the same blood proteins used to calculate biological age, the team cannot yet determine whether the observed changes stem directly from an anti‑aging effect or simply reflect improvements in lung function.

The study lasted 12 weeks, and the researchers noted that the drug remains investigational and is not approved for anti‑aging use. Side effects reported so far were mild, including occasional diarrhea, slightly reduced potassium levels, and temporary elevations in liver enzymes, all of which returned to normal after discontinuation.

Next steps

Insilico Medicine plans a year‑long Phase 3 trial in China involving more than 300 IPF patients. That larger study will provide a clearer picture of rentosertib’s safety and efficacy for the lung disease and may shed additional light on the biological‑aging signals observed in the pilot.

Researchers also suggest future trials should include healthy older adults without IPF to determine whether the drug’s impact on aging markers extends beyond disease treatment.

Broader context

While the early data are intriguing, experts caution against drawing conclusions about real‑world health benefits. “The apparent age‑related changes were based on biological markers, not demonstrated changes in a person’s actual lifespan or overall health,” noted Insilico executive Dr. Carol Ann Satler.

Nevertheless, the study adds to a growing body of research exploring how targeted therapies might influence the biological processes of aging, a field that many see as a potential avenue for improving healthspan for all Americans.
