Alumis to Pursue Late‑Stage Lupus Trial After Mid‑Stage Setback

Alumis, a biotech firm headquartered in San Francisco that specializes in autoimmune‑disease therapies, said Tuesday it will move forward with a late‑stage clinical trial of its oral drug envudeucitinib for systemic lupus erythematosus (SLE). The decision comes after the company’s Phase II study failed to achieve its primary endpoint.

Mid‑stage trial results

The Phase II trial enrolled 408 adult participants with moderate‑to‑severe SLE and treated them for 48 weeks. Although the study did not meet the overall primary efficacy goal, the company highlighted a subgroup of patients who exhibited higher interferon activity – a biomarker linked to disease severity. In that subgroup, envudeucitinib produced a statistically significant improvement on the trial’s main disease‑activity measure.

Why the subgroup matters

Alumis explained that patients with elevated interferon signaling are thought to respond better to therapies that target the interferon pathway. Because this subgroup represented a smaller portion of the trial population than anticipated, the company believes the overall miss was driven by dilution of the signal rather than a lack of drug efficacy.

Next steps

Based on these findings, Alumis plans to discuss a focused late‑stage (Phase III) trial with U.S. regulators, concentrating on patients with the high‑interferon profile. The company also noted that envudeucitinib was generally well tolerated, with no new safety concerns emerging during the study.

Broader context

Systemic lupus erythematosus affects an estimated 200,000 Americans, according to the Centers for Disease Control and Prevention. Effective treatments remain limited, and the disease has long been described as a “graveyard” for drug development. Alumis’ approach – targeting the TYK2 protein to calm overactive inflammation – reflects a growing strategy of precision medicine in autoimmune disorders.

Other pipeline activity

In addition to its lupus program, Alumis confirmed it remains on schedule to file a U.S. regulatory application for envudeucitinib in moderate‑to‑severe plaque psoriasis in the fourth quarter of 2026, following positive results from earlier trials.

Market reaction

Shares of Alumis fell more than 55 % in pre‑market trading after the news, as investors had viewed the mid‑stage study as a pivotal test for the company’s lupus ambitions.

While the setback underscores the challenges of developing new therapies for complex autoimmune diseases, Alumis’ focus on a biologically defined patient subgroup may offer a path forward for both the company and patients seeking better treatment options.
